[Abstract]
Breast cancer is a kind of malignant tumour and is common in female in our country. Its mortality is secondly to lung cancer, and incidences of breast cancer have a trend of rising straightly. The latest data from China Anticancer Association, which indicate the morality of women breast cancer in China is raising by 3 percent, breast cancer has became the first killer of female health.Taxol is one of the most effective treating breast cancer medicines, single or combine with other medicine also show well curative effect. Taxol have been used in clinic therapy of breast cancer. Generally speaking, taxol can accelerate the-assemble of microtubule, stabilize microtubule from its discomposing, make mitosis stagnate at G2/M, sequentially interdict the normal mitosis and proliferation of cells, induce cell apoptosis; Besides, a direct function of taxol on cell signal and gene expression, activations of apoptosis genes and expression of proteins can induce apoptosis. However, the molecular mechanism of taxol induce tumour cell apoptosis is not clear, moreover, the function of Taxol differ with different cell types.In this experiment, we use breast cancer MCF-7 cells cultured in vitro, after treatment of different time and concentration of taxol, chromosome DNA ladder electrophoresis was used to examine the degree of apoptosis; RT-PCR and Western blot were used to detect expression changes of apoptosis genes mRNA level and corresponding proteins in Fas/FasL pathway. In this study, we try to find the relation between breast cancer cells induced by taxol and Fas/FasL pathway, it can make a further indication of molecular mechanism of breast cancer cells apoptosis induced by taxol by providing experimental evidences. Experimental results show:1?Results of DNA ladder electrophoresis reveal that taxol can induce apoptosis in breast cells, and it can be along with time and concentration increased aggravating, indicate that apoptosis of MCF-7 cells induced by Taxol have a time and dose-dependent manner.2?Results of PCR reveal : in MCF-7 cells, when the concentration of taxol increased , the mRNA levels of gene Fas?FasL?FADD?Caspase-8?Caspase-10?Caspase-3 have a upgrade, while the mRNA levels of gene DR4?DR5 have no obvious changes.3?Results of Western-blot reveal: along with the increase concentration of taxol, the expressions of Fas?FasL proteins increased gradually, while the expressions of Caspase-8?Caspase-3 zymogen proteins decreased gradually.we can conclude though our results that cell apoptosis of human breast cancer MCF-7 cells induced by taxol relate to Fas/FasL pathway, while DR4?DR5 have not involved in this process.
